To do this it used to be you had to create a mini web page and save it as a web archive, then find the webarchives for mail signatures in the mail folder and overwrite one....not anymore

Its always good advice to back up your work and EMCs Retrospect is one of the premier tools to do this on a mac, though now there are a few more than before, including Leopards time machine.

The first thing to do is restart your mac to see if the problem goes away.

You can also restart with the shift key depressed and this starts your mac in safe mode, if things are OK now it means something you installed or added has caused a problem
